 MS culture medium was designed by Murashige and Skoog for tobacco cell culture in 1962. It is characterized by high concentration of inorganic salts and ions, and is a relatively stable ion balance solution. Its nitrate content is high, and the amount and proportion of nutrients are appropriate, which can meet the nutritional and physiological needs of plant cells. Therefore, it is widely used as the basic medium for rapid propagation of most plant tissue culture. Based on this, this culture medium was named after them.

 

MS medium is a very commonly used tissue culture medium, in which many variants have been developed. This culture medium originated from White's culture medium and was initially developed for the cultivation of tobacco callus tissue. Compared to white culture medium, the concentration of all components increased. Increasing nitrogen to 50-60mM significantly stimulates the growth of tobacco cells, but concentrations of 80mM or higher are clearly detrimental to the cells. The increase of all other elements, especially macro elements, also stimulates the growth of callus tissue. Due to the high concentration of minerals, MS medium is a very rich saline medium, which may be too salty for certain plant species. To avoid this issue, MS is usually used in conjunction with trace elements at full concentrations, but with constant elements at concentrations of half or three-quarters as originally described by the author. Sometimes, the original MS vitamins are replaced by vitamins from Linsmaier and Gamborg B5 culture media, as the concentration of thiamine is higher compared to the demand for this vitamin in plants.
